Antibody fragments, especially Fabs, are getting increased attention as potential biopharmaceutical agents.
Fabs usually retain the specific, antigen-binding affinity of the parent IgG. Such fragments have some advantages over monoclonal antibodies (Mabs), eg. they show improved pharmacokinetics for tissue penetration and they can bind to targets inaccessible to conventional antigen-binding sites.
To enable the use of Fabs in clinical applications, an efficient and high quality production process is needed, including the downstream purification process. This process needs to be robust, with high yields, high purity, and low leakage of ligand.
